Jane Applegate is enumerated with William Henry in the 1850 Census. They are residing in the household of John and Emaline A Innes. In his will, William identifies Emaline as his sister-in-law. Since I believe Jane is the wife of William, it seems probable she is the sister of either John Innes or his wife Emaline.
